The microscope in cell studies — common mistakes

Always convert all measurements to the same unit (e.g., micrometres, µm) before performing magnification calculations. A common mistake is forgetting this step. For Paper 2, show your working clearly; even if your final answer is wrong, you might earn method marks for correct unit conversion or formula rearrangement.

Magnification is how much larger an image appears compared to its actual size. Resolution, on the other hand, is the ability to distinguish between two separate points. A high-resolution image shows fine detail clearly, while a highly magnified but low-resolution image will appear blurry. Biologists use different microscopes because each type has distinct advantages and limitations. Light microscopes are simpler, cheaper, and allow for the observation of living specimens, but have lower magnification and resolution. Electron microscopes (like TEMs and SEMs) offer vastly superior magnification and resolution, revealing intricate ultrastructure, but they are expensive, complex, and require dead specimens. The choice depends on what details a biologist needs to observe.

To correctly calculate magnification or actual size using the formula M = I/A (Magnification = Image size / Actual size), you must first ensure that both the image size and actual size are in the same units (e.g., both in millimetres or both in micrometres). First, you calibrate the eyepiece graticule (which has arbitrary units) by lining it up with a stage micrometer (which has a known scale, e.g., 1 division = 10 µm). This allows you to calculate the actual size corresponding to one eyepiece unit at that specific magnification. Then, you can remove the stage micrometer, place your specimen on the stage, measure it in eyepiece units, and convert that measurement to an actual size (e.g., in µm).
