Testing for biological molecules — common mistakes

Cambridge examiners expect precise language. When describing results, always mention the initial colour and the final colour, along with any precipitates or emulsions. For example, 'the solution changed from blue to a brick-red precipitate' is far better than 'it turns red'.

Heating provides the activation energy for the redox reaction to occur between the reducing sugar and the copper(II) ions in Benedict's solution. Without sufficient heat (ideally from a water bath at >80°C), the characteristic colour change and brick-red precipitate indicating the presence of reducing sugars will not form, or will form very slowly. The key difference is the pre-treatment for non-reducing sugars. Reducing sugars react directly with Benedict's solution upon heating. Non-reducing sugars, like sucrose, first need to be hydrolysed (broken down) into their constituent reducing monosaccharides using dilute acid and heat, followed by neutralisation with alkali, before the Benedict's test can be performed to show a positive result. Focus on precision. Always state both initial and final colours, and differentiate between a solution, precipitate, and emulsion. Ensure correct temperatures (e.g., heating for Benedict's, room temperature for iodine). For non-reducing sugars, don't forget the neutralisation step.
